Now I am trying to download latest version of Telerik ASP.NET AJAX.
Which file should I download: Dev.zip or Dev.msi? Or both?

The Telerik_UI_for_ASP.NET_AJAX_2020_2_617_Dev_hotfix.zip zip which you can download from https://www.telerik.com/account/product-download?product=RCAJAX contains only the files you need to upgrade to the latest version.
The version of the archive name is 2020.2.617 which corresponds to R2 2020.

I just check my .DLL files and I have 2019 version so I still have to download only hotfix.zip file?

You should be able to access the latest version from your account.
Nevertheless, to save your precious time, please find attached the latest build to my reply.
The Bin40 folder contains the assemblies built against .NET 4.0, while the Bin45 folder is for .NET 4.5 and above.

Hi Vitaly,
Yes, in short, you need to replace the Telerik.Web.UI.dll and Telerik.Web.UI.Skins.dll files with the ones from the Bin40 or Bin45 folders depending on the .NET version of the project. If it is a web application, you also need to upgrade the references to them. All of this is explained with steps in the Manual Upgrade article.
Please don't forget to back up your project before starting the upgrade procedure.
Before starting an upgrade of your project, you may find it useful to review the following resources:
- The Known Issues and Important Changes sticky thread. It lists changes that may affect your code and known regression issues (that are usually fixed in the subsequent release). Review the information for the releases you go through so you can evaluate whether you will be affected by anything.
- The Telerik Upgrade API Analyzer can analyze your C# code to notify you of changes between your version and the new version you want to upgrade to. It offers information as of the Q1 2012 (2012.1.225) release.
- UI for ASP.NET AJAX Release History—reviewing the release notes for all releases you go through will let you see what has changed, what fixes, features and controls have been implemented so you are better prepared to meet your project's challenges.

I downloaded .dll files and copied to my bin folder however I am getting error on build:
Error 'Could not load file or assembly 'Telerik.Web.UI, Version=2020.2.617.40, Culture=neutral, PublicKeyToken=121fae78165ba3d4' or one of its dependencies. Operation is not supported. (Exception from HRESULT: 0x80131515)'

To fix it clean the contents of the licenses.licx file, save it and rebuild the app.

I removed license file but now I am still getting error on rebuild:

Can you please create a new empty license.licx and rebuild the app?
If this does not help:
- remove the reference to Telerik.Web.UI.dll in Visual Studio references tand re-add it.
- clean the Temporary ASP.NET Files folder - https://stackoverflow.com/questions/16137457/asp-net-temporary-files-cleanup
- clean Visual Studio cache - https://errorhandlinginskills.wordpress.com/2018/07/28/how-to-clear-visual-studio-cache/
- restart the PC
- and test again the project

It is working on local machine but when I moved to development server these files and try to test I am getting again the same error:
