Activating Kendo UI License on Build Agent

Does this command need to be done for each build or once per build environment?

This depends on that if you are building the project locally, or you are building it using CI services.

If you are building the project locally, it is enough to activate the license once.

However, if you are building it using CI services, the license activation has to be one of the steps when deploying the application. Also, you have to ensure that the license activation on the CI is done in the same folder where the application is:
