Fiddler is failing set up the system proxy.
This is what the log says:

11:37:30:6321 Fiddler was unable to get information about the proxy for 'DefaultLAN' [0x2f92].
11:37:30:6321 !WARNING: Failed to get proxy information for Connection 'DefaultLAN'.
11:37:30:6321 Setting upstream gateway to none
11:37:30:6351 Windows 8+ AppContainer isolation feature detected.
I also tried to manually change the proxy settings from inetcpl.cpl. but when I try to click "OK" in the "LAN Settings" dialog, nothing happens. The "Cancel" button does work though.

The problem doesn't seem to be Fiddler related. For some reason you cannot change the proxy settings on your machine. That has to be fixed for Fiddler to work. Is it possible that this is domain policy?

Also receiving the error with windows 10 creators update. It looks like fiddler is not compatible with Windows 10 creators update build 1703?

That is exactly what I am experiencing as well. I'm guessing Microsoft may have broken something with IE 11 in the latest Windows 10 Creators Update. Hopefully Telerik is on the "red phone" with Microsoft to let them know about this issue as I am sure they have better sway then I would. I also hope the telerik folks are able to replicate this issue.
We've created a virtual machine with fresh Win 10 Creators update installation and there was no problem whatsoever. This makes me think that the update procedure have broken something. There are other people over the Internet that experience the same problem like yours. Also there are some tips and tricks to try to fix this behavior, but be careful since most (if not all) of them are not official troubleshooting guides from Microsoft and you are using them at your own risk.

What info on the internet did you find that might fix this issue? I so far have found nothing that hints at a solution to this problem.
Also, is there a debug flag to turn on fiddler that might yield more information as to why fiddler gets this error:

Rather unfortunately, in this case turning debugging logging on will not yield more information.

Hopefully Telerik will update fiddler to detect if that service is stopped and notify the user to enable it.

In debug mode, fiddler could be set as system proxy successful;
but in release mode, failed to register Fiddler as the system proxy.
and WinHTTP Web Proxy Auto-Discovery Service is opened.

I am not entirely certain what is meant by debug or release mode. Is it possible to provide the below information on this scenario?
1. The output of the Fiddler Log Tab.
2. A Session Archive Zip of the session in both debug and release mode.

This warning happens to me when I try to use https://www.telerik.com/forums/download-stand-alone-version-of-fiddler
This warning does not happen to me when I run a full installation.
Does it help anything?
