Hello James,
After the Q2 2010 release the RadTreeListView inherits the GridViewDataControl, not the TreeView.
It is in our plans to bring the tree lines back, but it will not be easy and I cannot promise any specific time-frame.

Are tree lines still not supported?
Are there any other tricks we can use to allow the treeview heirachy to be better visualised? Is there a way to add a leading line that goes to the root node? or colouring different nodes sections?

This feature has not yet been implemented and for the time being is not on our schedule.
However you are free to re- template, and restyle the TreeList rows any way you like, so you can achieve almost any look you want.
