I don't quite understand your question.
If you mean a possibility to assign properties of the ExtendedData after the PreviewReadComplete, then yes.But when you change whole ExtendedData after the PreviewReadComplete, then the binding will be corrupted. For example the tooltip which is assigned via MapShapeReader.ToolTipFormat property will not work.
